Synthesis
The DNA Analysis Facility offers the synthesis of oligonucleotides in 2 scales. Synthesis turnaround times are typically 24 to 48 hours. All oligos are quantitated by A260 and analyzed for hairpin and dimer formation. This information is available to the user on the JHU Finch Server.
50nmol
The smallest scale, the 50 nmol scale, produces a minimum concentration of 2.5 µM in 750µl of 20% acetonitrile (ACN) up to a maximum concentration of 10.0 µM in 750µl (1.8 nmol to 7.5 nmol). The oligos produced at this scale are purified automatically to remove any "N-minus" synthesis byproducts and are returned to the customer in 20% ACN.
0.2 µmol
The next larger scale is the 0.2 µmol scale. Typical yields for this scale are 1.0ml at 50 to 112 µM. Post-synthesis purification is available at an additional charge and must be selected before synthesis starts. After synthesis at the 0.2 µmol scale the oligos are desalted over a Sephadex G25 column to reduce the pH back to 7.1 to 7.6 and returned to the customer in sterile deionized water.
